here's my list of important guitarists, in no particular order (I am thin on classical music, so ...)
- Jimi Hendrix - where would we all be without him and "live at monterey"
- Eddie Van Halen - not that he's "best", but he promoted guitar during 80's like nobody else.
- Dave Murray (and Adrian Smith) - apart from being underrated kickass guitar player (he was tapping and doing hammer-on solos way before Van Halen learned how to do it), they kinda set the sound and theme for the whole slew of heavy metal bands back then. Some may have played more distorted or faster, but they were just repeating what these 2 did.
- Tony Iommy - for the "paranoid" riff (aka you don't have to play like god to be a guitar hero), it gave hope to everyone fiddling with guitar 
- Angus Young - can you say "riff"? For the greatest riffs ever (that have been copied bazillion times), we thank you!
- Joe Satriani & Steve Vai - while I outgrew their particular style of music and composition, you cannot argue that these two moved the limit of what was thought possible on electric guitar. If you are guitar freak, you probably find them amazing, if you are not, you probably don't get it.
- Michael Hedges - Jimi Hendrix of acoustic guitar. Before him, people played either country/folk, or classical/flamenco. Hi grabbed his Martin and did something else. He insipired a number of artists today (like Ani DeFranco) who are basically repeating his licks. Also scores cool points for playing in odd tunings and on weird instruments (harp guitar).
- Paco De Lucia - There are probably better flamenco guitarists out there, but he's the greatest imo. The first acoustic guitar LP I ever owned was Paco. (In addition to him, you gotta add Al DiMeola and John McLaughlin for making that San Francisco trio concert, which showed that you don't need electric guitars to be virtuoso.)
- Randy Rhodes & Zakk Wylde - I only listened to these two playing with Ozzy, but man they could kick ass. Ok, Steve Vai has more licks, but I'd chose Ozzy cd anytime before I chose Vai.
- David Gilmour - the guy with best feel ever, hands down. If you ever watched Pink Floyd live, you know what I'm talking about. If you haven't you missed something epic.
